What was actually found

Leptobacillium doupengense was isolated from humus-rich material accumulated in a tree hollow in the Doupengshan area near Duyun, in the Chinese province of Guizhou. The region has a karst landscape in which small pockets of organic matter can form distinct microenvironments. The specific name alludes to the origin of the material.

Unlike showy cap fungi, this taxon is known above all from culture. The authors described the rate and appearance of colonies on media, the structure of the hyphae, the conidiogenous cells and the shape of the asexual spores. The characters were set against descriptions of known Leptobacillium species. Analyses of DNA sequences placed the isolates within the genus, but in a distinct, well-supported lineage.

The humus of a tree hollow is not simply "soil in a tree". It retains water, fragments of leaves, droppings and decaying wood, while being isolated from the surface soil. For microfungi it may constitute a stable island of resources. Isolation alone does not, however, explain the function of L. doupengense: the publication does not establish whether in nature it is primarily a saprotroph, an endophyte, a parasite of other fungi, or an organism using several strategies.

Isolation from a sample also calls for careful interpretation. What grows on a plate may be an organism abundant in the environment, but equally a single spore carried in on the air. Repeatable isolates, observation directly in the material and data from many hollows would allow a check on whether it is a constant component of this microhabitat. Success in culture is not a measure of its abundance in nature.

A description of a culture should be repeatable under the same conditions. Temperature, medium composition and incubation time change the diameter, texture and colour of a colony. Without those parameters, comparison between two laboratories loses value. The authors state the conditions so that an image does not function as a detached illustration. Such standardisation makes it possible to distinguish a character of the species from a response to culture.

That distinction matters, because genera containing similar anamorphs can be ecologically diverse. Naming a species creates a tool for further experiments; it does not close its biology in a single sentence. A safeguarded culture allows observations to be repeated and future isolates from other hollows and karst habitats to be compared. Only a larger sample will show the range and ecological significance of the new taxon.

What the paper establishes

Leptobacillium doupengense (MB 862246) was described from two cultures isolated from humus in holes of unidentified trees at Doupengshan, Guizhou. The holotype is dried culture GZUIFR DPS001; ex-type culture GZUIFR DPS003 derives from it. Five loci—ITS, nrSSU, nrLSU, tef1-α and rpb1—formed a lineage with 100% ML/1 PP support. Slender chains of 3–4 × 0.5–1 µm conidia and colonies reaching 35–40 mm after 14 days at 25°C also distinguish it. Recovery from tree-hole humus does not prove it is a persistent cavity resident, a tree pathogen or an insect pathogen; direct observation and repeated isolation are needed. Two cultures from one area cannot establish its range.
